Antioxidant Activity of Backhousia citriodora and Its Effects on Osteogenesis in Artificially-Induced Menopausal Rats

Abstract
This study was designed to investigate the antioxidant activity of Backhousia citriodora (BC) and its effect on osteogenesis in artificially-induced menopausal rats. The antioxidant activity of BC was evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity assay and superoxide dismutase (SOD)-like activity assay. BC extracts showed similar DPPH radical scavenging activity as ascorbic acid, which was used as a positive control at a concentration over 50 ¥ìg/mL. The SOD-like activities of BC extracts increased in a concentration-dependent manner. Especially, SOD activity of BC ethanol (BCE) extract at a concentration of 500¡­1,000 ¥ìg/mL was higher than that of hot water extract. In addition, total polyphenol concentration of BCE extract was higher than that of hot water extract. Therefore, BCE extract was used for animal experiments. In order to investigate the effects of BCE extract on osteogenesis in artificially-induced menopausal rats, 9-week-old female Sprague-Dawley rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-CON) and ovariectomized rats supplemented with BCE extract at 50 mg/kg bw and 200 mg/kg bw, respectively. Serum alkaline phosphatase activity, an indicators of osteogenesis, of the BC extract group was lower than that of the OVX-CON group. Collagen contents in cartilage, bone, and skin were reduced by ovariectomy but increased upon BC treatment, especially in the 50 mg/kg bw group. According to these results, BC extract had improvement effects on osteogenesis in ovariectomized rats.
